昨天講好說一定要寫這麼一篇文章的，

結果我又很早就跑去睡覺，忘了。

話說，關於我那堆接了老半天都接不出來的construct，

直到上周進度大概是剩下最後兩個，

N-terminal GFP CEN、C-terminal GFP 2μ，

也是屬於struggle很久的類型，

第一個困難點是subclone前原本的insert和vector size差異只有幾百個bp，

在agarose gel很難分開；

第二個construct是，除了第一個的問題之外，是我好久沒做的3-fragment ligation，

好啦～我個人對於construct的態度一直都是埋頭苦幹實幹，總有一天會是我的，從小就降子。。。

總之就是降子，從九月做到現在，換了許多方法。

卻，敵不上我們家小開的一句話。

這禮拜佩秦發現，學妹接好的C-terminal HA好像比之前的N-terminal HA還要punctuate，

其實這時候大概就知道N-terminal GFP的constrcut可以不要接了，

只是，我實在是不喜歡事情做一半，尤其是，都接好了只剩下挑transformant做miniprep，

幹嘛不做啊！

昨天miniprep完，正在做enzyme digestion check的時間，

去開了group meeting，討論了一下實驗，

老師是很明確的講說，那個N-terminal GFP就不要做了。。。

你看你看，一定是因為他這句話，

昨天miniprep的結果，雖然最近又有某些不知明原因讓我的plasmid抽出來的量很少，

24隻裡面，只有9隻的DNA是可以明顯的在agarose gel上看到，

其中，有3隻是vector alone，剩下的通通是！

很久沒這種中獎率這麼高的經驗了！

老師還說叫我騙我的本子，

結果，他講一句話比我在本子上亂寫一百句都有用吧，

只要說你不想要這個data就好了啊。。。
